EBAE II: Electric Boogaloo

Woot! This past week’s meeting, EBAE II, was a great success! It was a great discussion and relaying of some useful tips and tricks. Many thanks to Koji Takahashi and Forrest Collman for their contributions. And thanks to the SUPD Postdoc Initiative Fund for funds to cover dinner.

Some folks from the Deisseroth lab have provided recipes for NMDG slicing solutions. They provided some useful details about them:

Here are receipes for the NMDG cutting solution and modified HEPES ACSF for holding slices until recording. The procedure is to perfuse and slice with NMDG, leave the slices in NMDG for 5-10 min (apparently the optimal time to leave your slices in NMDG is somewhat brain area dependent…)  at 33deg, then do 3 x 10 min washes with the HEPES ACSF before starting recording. The NMDG solution is a huge pain in the ass. There’s a bunch of stuff that needs to get made up fresh each day and apparently the NMDG only has a useful shelf life of ~3wks, so you have to keep reordering.
